Rucinol
Rucinol is a derivative of resorcinol found in fir trees in Siberia. It works by inhibiting the activity of tyrosinase and tyrosinase-related protein-1. One randomized controlled trial showed Rucinol serum 0.3% to have efficacy in treating melasma as evidenced by significantly lower clinical pigmentation scores after 12 weeks.17 Liposome encapsulation has been shown to improve the stabilization and penetration of Rucinol.21
Methimazole Methimazole is an emerging agent used for depigmentation. It is a peroxidase inhibitor that is a noncytotoxic inhibitor of melanin production. The first case report of topical 5% methimazole use in treating PIH in a patient showed a moderate improvement of the hyperpigmented areas following daily use for 6 weeks.22 Another report showed significant improvement of melasma in HQ-resistant melasma patients using the application of 5% methimazole cream once daily for 8 weeks.23
