Cultured epidermal suspensions

The CES technique involves the culturing of both melanocytes and keratinocytes. Their dual presence is beneficial in that they can organize in a manner mirroring the basal layer of the skin. Using this method, an ultra-thin skin graft is harvested and trypsinized. The isolated melanocytes and keratinocytes are placed in culture media containing growth factors for both cell types for a few weeks. Dispase can then be used to detach the formed epidermal sheet, which is subsequently placed on top of paraffin gauze and transferred to the prepared RS. When compared to CMS, less culture time is required, though like CMS this is a costly procedure (Fig. 52-7).12,54
